Global public health and Structural & Protein Sciences departments have a new opportunity for a dedicated, highly motivated Post-Doctoral Fellow with an interest in viral protein structural biology (X-Ray Crystallography/Cryo-EM) to investigate Coronavirus proteins for the fight against SARS-CoV-2 and other future coronavirus epidemics. GPH is committed to addressing the medical needs of patients in the developing world and resource limited settings and utilizes the resources, capabilities, pharmaceuticals, technologies, access programs, policy initiatives and programs of JRD to positively impact the health and well-being of underserved communities globally. The GPH portfolio includes projects in Coronavirus, Dengue, Tuberculosis, HIV treatment and a Long-Acting formulation platform for HIV, TB, Dengue and Malaria. The Structural & Protein Sciences (S&PS) department is a global and coordinated team that applies state-of-the-art capabilities to enable small molecule discovery for Janssen R&D by generating high quality proteins to enable assay development, HTS, pharmacological profiling and structural biology (X-ray crystallography, Cryo-EM and NMR). The Protein Science Team collaborates closely with Janssen Therapeutic Areas and functional teams and with external collaborators in academia and industry. The postdoctoral project will entail design, engineering, and production of various coronavirus structural and non-structural proteins and employ structural biology to investigate the functional implications. You will be working at our state-of the art protein science laboratories in Spring House, PA. The successful candidate will be playing a key role in supporting our efforts in understanding the coronavirus assembly, but also with the potential to work on other Neglected Tropical Diseases. Responsibilities: Construct design, expression screening, and production of various coronaviral structural and non-structural proteins. Biophysical and biochemical characterization of recombinant proteins. Perform X-ray crystallography and/or Cryo-EM methods to investigate the coronavirus assembly and function. Engaging with GPH and S&PS teams and leverage innovative laboratory technology/infrastructure. Working towards publishing in high-impact journals and to present your research at internal and external meetings. Qualifications PhD in Biological Sciences, Virology, Biochemistry, Structural Biology or related fields. Experience in construct design and purification of proteins for structure determination using standard techniques and equipment (ex. Westerns, SDS-PAGE, affinity, ion exchange, size exclusion chromatography via AKTA) is required. Experience in protein characterization using techniques (such as analytical gel filtration, mass spectrometry, NanoDSF, and/or dynamic light scattering) is preferred. Experience in membrane protein expression and purification is preferred. Hands-on experience with single particle cryo-EM sample preparation, optimization, data collection and processing is preferred. Experience with target deconvolution and the analysis of complex data sets is preferred. Demonstrate a sense of urgency, be organized and adhere to timelines. Excellent written, oral, and interpersonal communication skills. Excellent problem-solving skills and the competence to adapt to changing priorities. Ability to operate effectively in a team setting within a multicultural environment.
