Only take the job if the compensation is fair, your manager is fair and it fits within your overall lifestyle (need more training, get out a contract role into permanent, etc.) But do not plan on staying here until retirement, the culture is toxic.

Depends on the department, but mostly be aware of the relative inflexibility and general belief that nobody should be allowed to work from home or other remote location unless it specifically involves work=related travel; i.e., a business meeting.
