Preschool and early schooling is probably the best time to relocate with children. 2 years isn't forever, but you'll be NETWORKING the whole time with people who you can let know that you'll be available after this engagement is up. You're already planning to rent, not sell the house, so you'll have a place to go back to and rentals are still hot on the market.
I don't see any reasons NOT to go for it other than the medical/physical aspects and your spouse's job. It would definitely be an adjustment, but somewhere that wants you will absolutely, 100% work with you on your medical stuff. My cousin had kidney failure in his early 20's due to a genetic condition and throughout hiring at high end companies, his medical was accommodated for. When they want YOU, they'll make it work.
